Hi everyone,
we are having few problems in setting up a technique to combine DAKO
myeloperoxidase MoAb recognition with surface antigens by flow in
leukemia. We are using (after suface MoAbs) a fixation procedure that
includes paraformaldeyde 1% for 15 minutes on ice and -20 degrees ETOH 70%
overnight.
The problem is that we are having positivity in negative samples.
We are also staining for Tdt, cytoplasmic CD22 and CD3. Is there any
fixation procedure that can be used for all of them?
Any suggestion will be appreciated.
Thanks for your help!
