re: labelling immature granulocytes

Having played with this a little, I can offer my approaches:
1. myeloid maturation can be viewed well with combinations of CD13, CD13,
and CD45 (nike swash pattern) or CD11b, CD16, CD45 (boomerang pattern). I
reccommend always doing both.
2. immature myeloid cells are CD32-, CD64+(dim) and with maturation CD64
is lost and CD32 is gained. The only caveat is that CD64 can be
upregulated on mature cells with infection or iatrogenic administration
of interferon-gamma, G-CSF, GM-CSF or IL-12.
3. CD33 I find of little additional value.

Beyond these suggestions, other information is available through a
literature review or some reading in the library. Suggest looking at work
of Loken and Terstappen, as well as becoming aware of the many antigenic
changes that occur with neutrophil activation.
