We do not use separation medium on single cell suspensions from lymph nodes
of
mice. FCS and SSC profiles normally show a single population of lymphocytes
only.

Can someone please tell me if there is any benefit to using density gradient

separation on cells from a lymph node (be it a biopsy or FNA) if you are
already using viability gating?
What I saw with viability gating looked pretty straight forward, and I
couldn't see why any separation media would be needed.
I would be interested in any and all experience people have had with the two

techniques.
