Here comes the help (hope so)!
I'm working in the group of Andreas Radbruch in Cologne together with Mario Assenmacher
and JŸrgen Schmitz. They have developed a method for intracellular staining of
cytokines and have much experience in this business. They have no problems with
high background staining. Here are some references from which you can get
information about their method and the results:

Schmitz, J., Assenmacher, M. and A. Radbruch. 1993. Regulation of Th cell
cytokine expression: functional dichotomy of APCs. Eur. J. Immunol. 23: 191

Assenmacher, M., Schmitz, J. and A. Radbruch. 1994. Flow cytometric determination
of cytokines in activated murine Th lymphocytes. Expression of IL-10 in
IFN-gamma and in IL-4 expressing cells. Eur. J. Immunol. 24: 1097

A. Radbruch. Flow Cytometry and Cell Sorting. Springer Laboratory. ISBN
0-387-55594-3

If you think it's interesting for you and you need more detailed information you
can send me your Fax-No. and I will send you the protocol.

Good Luck ;-)
