re: analysis for dim fluorescence

We have used the histogram subtraction technique with data from
a Cytofluorograf II. The software we used was Multi2D from Phoenix
Flow Systems. This software uses the methods of Overton (Cytometry,
1988 Modified histogram subtraction technique for analysis of flow
cytometry data. v9:619-626) to perform the subtraction and present
a percent of positive cells as the result.

Be aware, that all programs do not perform the subtraction the same
way. Multi2D calculates the percentage of cells with increased
probe incorporation. Other software programs just do a channel
by channel subtraction, which can lead to spurious results, and even
negative values.
