RBC counts in Turkeys

One of our scientists is interested in using flow cytometry to determine
absolute RBC counts in turkeys (the feathered kind). Our routine
hematology instruments will not differentiate the nucleated RBCs. A
FACScan and VANTAGE (soon) are available for the assay, but I would
prefer to use the former.

Are any pan leukocyte markers available for turkeys? Will the RBCs stain
differentially with DNA dyes? Has anyone used the Cytek absolute count
system with a FACScan?

To start with, I am thinking of the following experiment.

1. Fix peripheral blood with 1% glutaraldehyde/PBS, at 4 degrees C
overnight.

2. Aspirate fixative and resuspend in PBS with 30 Ug/ml SDS to sphere RBCs.

3. Run samples and look for light scatter clusters.

Am I anywhere near the ball park here? Thanks for your comments.
