WE WOULD LIKE TO STIMULATE HUMAN PBL'S WITH A SUPERANTIGEN (FOR EXAMPLE
STAFF. AUREUS TOXIC SCHOCK SYNDROME TOXIN-1;TSST-1 FROM TOXIN TECHNOLOGY)
AS A CONTROL IN ANALYSING TCR-V-BETA USAGE IN ANTIGEN STIMULATION TESTS.
THE IDEA IS TO LOOK FOR CHANGES IN FREQUENCY OF TCR-V-BETA+ CELLS IN T
-CELL SUBSETS BEFORE AND AFTER STIMULATION.

SINCE WE HAVE PREVIOUSLY NOT USED SUPERANTIGENS IT WOULD BE NICE TO GET
SOME HINTS AS TO THE OPTIMAL CONDITIONS (CONCENTRATIONS, KINETICS) AND WHAT
THE BEST CONTROL FOR OPTIMAL STIMULATION COULD BE.
INITIALLY WE HAVEN'T SEEN MUCH STIMULATION USING 10 NG/ML/10(6)CELLS.
STIMULATION WAS FOR 4 HOURS AND AFTER WASHING THE CELLS WERE CULTURED FOR
72 HOURS WITHOUT IL-2. AFTER WASH, IL-2 (1IU/ML) WAS ADDED FOR 24 HOURS.

ANY SUGGESTION WHAT MAY BE WRONG??
