Hi Tom. We have been routinely using an EPICS Elite with a two-laser
configuration to measure calcium uptake of T-hybridoma cells using
Indo-1AM loading. Both lasers are air cooled;the argon is 15 mw and the
UV source is an Omnichrome HeCd air-cooled 20 mw. We use the HeCd to
measure the short wavelength (a 440 Dichroic with a 385 band pass) and
the argon to measure the long (a 550 dichroic with a 525 band pass).
This system utilizes the gated amp configuration with the two lasers
being non-colinear (wow what a phrase). We were somewhat concerned
initially about the possible "noisy" signals of the HeCd, but much to our
surprise and delight, we get excellent calcium "rise" histograms using
this set-up. Additionally, we use this HeCd for DNA analyses and have
been quite pleased with the results. I would be happy to give you the
complete details (filter set up, etc.) if you are interested.
