Greetings from downunder,

At QIMR we do a lot of different experiments with Calcium Flux using INDO-1
as well as work with other UV excitable probes such as Hoechst 33342 &
33256, MNA. Do you use pulse processing for your Calcium Flux work? Also
what filters do you use? We use pulse processing ie the ratio option which
is much more efficient at showing calcium changes than just the individual
violet and blue colours. Filters used are a 405/20 for the violet and 530/30
for the blue with a 480 LP dichroic.
We have a FACS Vantage and our UV source is a Coherent Innova-90.5 Argon
laser. We use 50mW of UV for most of our UV work and find this is
sufficient.
We have all Coherent Lasers in our lab including a dye laser which we are
happy with. It is better to stick with one company for you lasers rather
than having a mixture of them.
