CD14,myeloperoxidase

Greetings to the Flow world,
I have two unrelated issues to which I am seeking guidance. #1 In our
acute myeloid leukemia panel we run two CD14 markers; MY4 and Mo2.
Recently, we had an M5 acute monocytic leukemia who was negative for both
these markers (confirmed by a positive alpha-napthyl butyrate and
negative sudan black). We have noticed that these markers are often
inconsistent in M5's and AMML's which is not unexpected. We also run a
CD36 as an additional mono/megakaryocytic marker. I am asking for other
investigators' input on what CD14 (or other) marker(s) would be best in
acute monocytic/myelomonocytic leukemias?
#2 I am looking for guidance for whole blood, myeloperoxidase staining by
Flow. We have been working with a FITC conjugated anti-MPO but am not
happy with non-specific fluorescence in FL2 which makes it difficult to
evaluate dual expression of surface antigens using PE labelled
antibodies. Any technical or other recommendations would be greatly
appreciated.
Thanks in advance,
Lori Rudzitis
