Dear David,

Many years ago (Exp. Cell Res. 80, 105-110, 1973) Dale Holm and I made the
measurement you are asking about. Obviously the answer depends on the
microspheres used and the instrument. At that time we were measuring CV's
on lymphocytes of about 3-4% and microspheres were coming up with a CV of
more like 3% or a little less. Our measurments indicated that about half
the CV was due to true bead variability and half due to our flow cytometry
measurement. More recently Dick Keller, Jim Jett, and John Martin (and
others) at Los Alamos have been using their high sensitivity instrument and
very small microspheres to repeat this measurement. With their system and
the spheres used they were coming up with the same results - half and half.
