Hello!
I am relatively new to sorting. We use a FACSStar machine. When I test
recovery using standard beads, I get good recovery based on a slide test.
However,under the same conditions when I try to collect myeloma cells in
G1 phase (PI staining) by DNA staining the recovery is very poor, even though
I collect upto a million cells for DNA extraction.
I use glass tubes for collecting the sorted cells, i.e. I think the glass tubes
do not pick up a charge as do plastic tubes.

What might be the problem(s)?
