stimulation of monocytes: fresh versus frozen

Hello everybody,
we start here in our department a clinical trial
and have to investigate the activation of monocytes
by analysing intracellular cytokine production.

Some times ago, I had a similar question about
CD-marker phenotyping, but now, in contrast,
we want to make functional assays.

Therefore, I want to specify my questions:

1) How do you handle the storage / freezing:
	Is it possible to store whole blood or only
	the PBMCs?
	Do you use DMSO or glycerin for storage?
	Which concentrations of DMSO?

2) Are the results (for example stimulation with
	LPA: fresh versus stored samples) comparable?
