markers to distinguish mouse monocytes

Hi, all!

In response to my earlier request for the best markers to phenotype human
monocytes, several people have asked the flip side of the question--the best
markers to identify  monocytes/macrophages in the mouse. Because there were
so many interested in mouse monocytes, I have decided to reply to the entire
group.

In our hands, CD11b (Mac-1) and Gr-1 (Ly-6G; clone RB6-8C5) best resolve
monocytes/macrophages from granulocytes in the spleen and blood of mice.
Both monocytes and granulocytes express CD11b but only granulocytes express
Gr-1. So, we routinely include both CD11b and Gr-1 in our stains when we
want to enumerate monocytes/macrophages in the spleen and blood. When we
phenotype mouse blood, we also include an antibody to CD45 to gate out
residual RBCs and platelets.
