Dear Flowers, Does anyone know of a protocol to stain specificaly a given plasmid used in transfection, in order to be able to follow its uptake by cells during different exposition times ? I have been using DAPI a lot and know it's specific for double stranded DNA (as is plasmid DNA) but was unable to find any staining protocol out there, and I have to admit that I fear some stain leakage and non specific signal if I use it ! If anyone know any staining procedure that can be used through flow cytometry and will remain specific to plasmid DNA used, it would be much appreciate.
