Hi John, I've been using this method a lot recently for S-phase determination in T-cell lines. It's quick, cheap and gives quite a clean prep. 1. Wash & spin cells in PBS or HBSS 2. Resuspend in 2ml absolute ethanol for 1 hour at 4 degrees 3. Wash and resuspend in PBS/HBSS with 10ug/ml RNAse A and 50ug/ml PI. Leave at room temperature in the dark for 30 minutes. Good luck Dave On Thu, 23 May 2002 14:54:04 -0500 jmgonzal@javeriana.edu.co wrote: > > Hi: > > I am interested in a good home made PI protocol for DNA content in cell > cultures. > > JOHN MARIO GONZALEZ M.D., Ph.D. > Facultad de Ciencias > Pontificia Universidad Javeriana > Fax 00 57 1 2850503 > Phone 00 57 1 3208320 Ext 4085 > > ---------------------------------------- David Corry Centre for Research in Biomedicine Faculty of Applied Sciences University of the West of England 0117 344 3397 Email: David.Corry@uwe.ac.uk
