Aki: The CPT tubes are great for clinical trials because the collection tube and the processing tube are the same (less chance for mix ups with the labels) and they require less skill than a ficoll prep. However, with the CPT tubes RBC contamination is a big problem and the yields are lower than with ficoll. If your lab has the skill and time to do the ficoll, I would go with that. For general staining, whole blood followed by a lyse/fix reagent works really well.
