We currently do this experiment on both the FACStar plus and the FACS
Vantage. The populations are usually very similar. The problem usually is
associated with the staining. We have found that the cell nomber and
concentration of dye are critical to generating good profiles. We stain
mouse BM at 1 million per ml with 4ug/ml Hoechst for 60 min. When we sort
on the Vantage at hagher rates, we concentrate the cells after staining to
3-5 million per ml. The profiles are identical.
