Georg-
We have had the same experience trying to visualize P-gp on the surface of
freshly isolated T cells. At best staining with P-gp specific MAbs results
in a (not very impressive) shift of the negative population. Two
explanations have been offered: P-gp expression is considerably lower in T
cells than in drug resistant tumor cells; MAb binding is dependent on the
steric configuration of P-gp in the membrane.

There are anti-P-gp antibodies conjugated to e.g. PE available from BD and
other distributors. Possibly you could use these. (Unfortunately we had
some major trouble when we tried the antibody from BD in a "quick and dirty
experiment". I donīt know why: No p-pg expression on our cell lines or a
bad lot of antibody?)
Best regards
Georg
