Georg- We have had the same experience trying to visualize P-gp on the surface of freshly isolated T cells. At best staining with P-gp specific MAbs results in a (not very impressive) shift of the negative population. Two explanations have been offered: P-gp expression is considerably lower in T cells than in drug resistant tumor cells; MAb binding is dependent on the steric configuration of P-gp in the membrane.
