We use a very rapid and efficient method. We dilute MNC (after ficoll) in PBS+1
mM EDTA + 0,5% HSA + 5% ACD in a 50 mL tube. Then we performe a 15 seconds
centifugations at 1680 g (300 RPM with our centrifuge). We repeat the procedure
once or twice. It as the same results than 150 g for 10 minutes and is much less
expensive than RosetteSep.
