Dear friends, I have recently noticed a strange occurrence with several different PE labelled Ab conjugates. The problem is a second, extremely bright (off the scale)population that bleeds into FL3 and sometimes FL4 parameters. It appears to be artifact of some sort but I don't know where it is coming from, it is 2x or more the intensity of the positive population. I commonly see this with NK1.1-PE and TCR beta-PE conjugates and some other cytokines too. I am sending an example of mouse spleen cells stained with TCR beta-PE conjugate for reference. Can someone please tell me what may be causing this and how I may get rid of it. (p.s this isn't the prettiest sample to begin with the cells were at room temp. most of the day before staining, but they are usually treated a little nicer before staining and still have the problem in question.)
