Intracellular IL-1â in monocytes

I currently work with intacellular IL-1â in monocytes. My main problem in evaluating the levels of stimulated and unstimulated IL-1â in peripheral blood, as I find quite hard to set the cursors in unstimulated samples without cutting the stimulated population in the middle. Also, I have noticed that there's a "shoulder" in unstimulated samples which seems that it might be a positive signal of IL1-â. Could that it be possible? Any ideas?
