We perform 34 positive selections on PB and bone marrow quite often. On a
good day our purity is better than 90%. The beads do not affect the FSCxSSC
of the recovered cells. To check the purity be sure to use an 34 antibody
to a different epitope. I believe the Miltenyi 34 kit uses an antibody to
HPCA-2 for selection, therefore, you should use an antibody to HPCA-1 to
check your purity.
