All, I am using biotinylated rat anti-mouse CD4 and CD8 from Pharmingen to do immunohistochemistry on frozen sections of Balb/c scid NCr mice from NCI. This was SUPPOSED to be a negative control, but these mice are staining positively in the spleen for CD4 and CD8. I have observed this in mice 7-12 weeks old. Other tissues like kidney are negative. These mice are also ab-TCR negative. When assayed by FACS these mice do not show the same large populations of CD4 or CD8 positive cells as with IHC. NCI doesn't seem to have any record of this occurring, and these mice should be too young to be "leaky" as has been reported elsewhere. Has anyone seen this before?
