Fluorescence Particle Question

Help.  I know this is non-flow, but I also know that someone has an answer.
We want to look at nano-particles, pm size range, in tissue slices.  The
particles are FITC labeled and we are interested in looking for accumulation
in various organs after IV injection.  We know that looking at individual
particles is out of the question and are really interested in the seeing
accumulation to a detectable level.  What would you recommend for tissue
prep.  We are thinking frozen OCT sections and/or paraffin sections.  How
can we handle the fixing of the tissue as not to destroy the FITC signal or
should we fix at all.  We would then analyze by fluorescence microscopy on
the slides.  We will also be running flow on the blood to see if the
particles are sticking to any WBCs.  Any thoughts you all may have on any of
this would be appreciated.
