Hi Group, Given the recent discussions on the formation of a bacterial flow cytometry group this posting will hopefully fall on helpful ears. I am trying to study the microbial ecology of the deep subsurface of the Wits basin in South Africa. I have collected 50-70C water samples from between 2 and 3.6km deep and believe it or not, FC and DNA staining reveals microbial counts of up to 10e5 cells per mil in some samples. I want to use FISH to gain an insight into the phylogeny of the samples and wondered if anyone has any preservation protocols for environmental samples prior to FISH and then FC analysis. Thanks in advance and I appreciate any feedback as I am a little isolated in terms of libraries or journals!!!
