Mark, there have been a few posts on this topic lately, rewind and recap at http://flowcyt.cyto.purdue.edu/ (the leftmost search field type in 45RO or something) In most of our normal controls CD45RO and RA are mutually exclusive ie. CD4++CD45RO++ plus CD4++CD45RA will be close to 100% of CD4++ and same for CD8. This is obtained using two separate tubes, 4/8/RO and 4/8/RA. Feeling curious, I stained RO and RA in the same tube - just because Master Yoederer says not to ;) - and in one or two of our unusual patients there were 20-40% dual positive CD45RA++/CD45RO++ (0% in normals). I wonder if someone would comment why exactly it is bad to stain CD45RO and RA in the same tube? Also, is there a transition period where naives become memory and they dually express? In periphery?
