Bunny, as you said, CD8 dims are mostly CD3- NK cells, all expressing CD45RA. I have yet to see any CD8dimCD3- CD45RO+ lymphs. For memory/naive issues I gate only on CD8 and CD4 bright lymphs first. I'll be happy to share a few dotplots/histograms of my analysies if you would like. I report the cells as CD45RO bright, CD8 bright. Since it sounds like you're concentrating on T cells, you might want to look at the magnetic CD3 separation. Although I would not personally think that being necessary as the distinction between CD8 dims and brights is very clear, even in TPA activated cells.
