Danni, Your problem is most likely a pressure problem with the sheath tank air pressure or the sample tube air pressure or both. Turn on your cytometer and boot your computer. Launch Cellquest, connect to the cytometer and open the status page. Note your laser power and current, as well as sample voltage. In the first five minutes, your laser power should be 15mW and your laser current should be approx. 7 amps. Prime the instrument and observe the fluid flow through your flow cell and the waste (orange) line of your waste tank. After the five minute warm up period, and when your sheath tank has pressurized, put the sample tube holder to the side and take a reading of the the sample voltage on your status page. It should be approx. 10.23 volts. Also put a sample tube with DI water on the instrument and run on high. Check the sample voltage. It should be approx. 5.9 volts. Change the flow rate to low and sample voltage should change to approx. 6.3 volts. With any pressure problem first ensure that your sheath tank has no leaks, and change your bal-seal.
