At the risk of crossing over the line into crass commercialization, I
would like to offer the following comments. I think those who have
avoided using PE-Cy5 conjugates because of this issue should know they
have a viable option.
The issue in the past was non-specific staining of human monocytes by
the PE-Cy5 complex on conjugates of both antibodies and isotype
controls. This problem has now been essentially eliminated and "before"
and "after" examples can be seen on our website (www.caltag.com). At
least one other commercial company also offers PE-Cy5 conjugates which
do not have the monocyte binding problem.

Just a quick questions. I have had trouble phrasing the search properly of
the e-mail discussions, but there was a discussion a few years ago
concerning high backgrounds for PE-Cy5 staining. (The samples are coming
up positive with Isotype staining - I know, Isotype staining might not be a
true negative, but that is a different discussion. :-)) Is there a solution
for the high background other than using some other fluorochrome like PerCp
which doesn't have this problem, as I understand it. Thank you in advance
for your help.
