minimum standards for cell types?

Dear group,

I wonder if there is a set of rules by which cells are defined in
flow.

For example "a monocyte will be defined as having forward and side
scatter values of such and such and will also be positive for CD33 or
14 as well as 45".

I am asking because I noticed a lot of application notes say
monocytes are defined by side scatter vs. CD14/33 gating and I just
don't feel comfortable with that, esp. in activated cells. I did some
playing around with different markers and there usually are a few
distinct populations based on the parameters above. Not to mention
Propidium Iodide.

Any thoughts will be appreciated,

maciej
