specific CD4/CD8 cells in HIV

Dear flowers,
Sorry for my English ,but I'll try to explain what we
need it. In Bulgaria we carry on antiretroviral
therapy(HAART) and in our laboratory we monitoring
some basic populations  and many other subpopulations.
But now we want to measure specific CD4 and CD8 cells.
We'll purchase  a BD kit  for this pupose: Fastimmune
Cytokine Sistem. Now there is three way to make this:
ELISPOT, tetramer complexes  and  this intracellular
cytokine  assay. In other to measure  specific CD4 and
CD8 cells we need an specific peptides/proteins. In
this kit there is no specific
antigens(peptides/proteins) for this purpose.
I have the following questions: I would be vary
grateful, if you gave me  theoretical articles not
only practical suggestions.
1. Is it possible to activate CD4 and CD8 cells in
whole blood with only specific peptide/protein w/o MHC
presenting cells? What about theoretical explanation?
Is there any differences b/w CD4 and CD8?
2. If it is possible Where we may purchase this
antigens?
