Subject:		Antibody Binding Capacity Problems




In our laboratory we are currently using Quantum-PE and Quantum Simply
Cellular kits, from Flow Cytometry Standards Corporation, to measure
Antibody Binding Capacity (ABC)of Fc receptors on human neutrophils.  We
are getting very high ABC values which are 3 to 4 times higher than the
values reported in the literature by standard methodologies.  According
to the technical representative at FCSC/Bangs Laboratories, we are doing
the quantitation correctly but the cells are just extremely bright.

My question is:
    Does anyone have experience using these products?  In particular,
antibodies labeled with PE and not FITC?

Thank you.  I would appreciate any suggestions that you may have.
