Megakaryocyte ploidy

Not directly a flow problem, but in case someone can help?

  A collegue of mine is interested in megakaryocyte ploidy, however he is
having difficulty maintaining long-term meg culture. By day 10, most
megakaryocytes form platelets thus few 8N or 16N are seen. Currently cord
blood CD34 pos cells are cultured in 10% BSA IMDM with 10% non-heparinised
cord serum, transferin and insulin. Cytokines used include MCDF (50 ng/ml),
SCF (50 ng/ml) and IL-6 (20 ng/ml).
Can anyone suggest an ideal culture environment fort long-term meg growth
and development.
