Since we were wrong about the use of OX-40L as a marker for activated rat macs, we are searching once more. Does anyone have any thoughts on the use of nitric oxide synthase for this purpose? Since NOS only increases upon activation, rather than going from - to +, would anti-NOS, if combined with an anti-mac antibody such as anti-ED1, be sufficiently specific to quantitate these cells? If so, does anyone know of a source other than Pharmingen for anti NOS that will react in the rat? Pharmingen sells it only as a FITC conjugate but the ED1 we have already purchased is a FITC conjugate and we would like to use it rather than having to buy additional ED1 in another form.
