MOre along this line to one up the Dot Plot...density plots are great for
recognizing small pops and close population centers

Dear Becky
I take it that your question is not about the identification of eosinophils
in whole
blood. It is whether the dot plot method is preferable to the histogram
method for
analysing expression of surface molecules expressed at low density.

I feel that your dot plot method is acceptable, as it approximates an
Overton Subtraction
result - see documentation for FCS Express (www.denovosoftware.com) or the
literature
for Roy Overtons publication of the method. The histogram method is
preferred by
traditionalists that are used to distinguishing CD4s from CD8s, and is not
at all
suitable for analysis of receptors expressed at low density. I would be
curious to
hear what the real pros have to say to this, though.
