Hi Viktor. I understand your considerations regarding the better choice to express your results. We have to deal with these questions sometimes. Sometimes we found that expressing data not only as % of gated lymphocytes or % of gated subpopulation, the information of absolute number of cells/mm3 of blood can be the best choice. It does not make me fell uncomfortable because in some situations different approaches can be the best tool. 

Regarding to the HLA-DR expression on CD8+ cells I have to tell you that you had the same problem in the past. We found out that the striking point was that we were analyzing HLA-DR expression only on CD8+ CD3+ cells (or CD8high+ or Bright+). This approach make the % of gated CD8+HLA-DR positive cells higher than the data presented in the literature because most of the published data in the past used only two color to discriminate CD8+HLA-DR+ cells. The major problem with this dual color analysis is that CD8low+ (probably NK cells does not express HLA-DR in the same proportion of CD8+ T cells.
