I can't help with the first, but for the second I got a lot out of Video
Microscopy by the legendary Shinya Inoue, and Digital Image Processing, by
Gregory Baxes.  The latter is a skinny thing, but very helpful.

For a solid introduction to both subjects I strongly suggest the Woods
Hole Microscopy course, probably starting next week (too late).  A word
of caution:  it's intense.  When they say "check in by 3pm", that's
because lectures start at 3:15, and you'll be in lab till 11pm!
