So far I haven't used a fixative for platelets that doesn't increase the base line for P-selectin expression. However, for studying relative levels, then fixation with 1% PFA for 5 mins at room temp. This will allow you to read the samples the next day. I would suggest you analyse the samples in citrated blood asap. Mody et al (1999) have found that using CTAD anticoagulated samples provide a longer period for analysis without in vitro activation. Of course reading Alan Michelson's reviews on analysis of platelet activation by flow cytometry will help you decide.
