When I needed a quick human PBMC sample to check whether an antibody had gone off or
something I would layer 0.5ml diluted blood onto 0.5ml ficol in a 1.5ml microcentrifuge
tube. I'd then spin it at 3000rpm for 5 minutes. I never optimized it for yield as I
didn't care if I lost some cells, seemed to work quite well though.
