Hello, just in the process of submitting a paper for publication we have the problem to explain a slightly curious phenomenon. A monoclonal towards a new receptor which we are characterizing when preincubated with the cells will inhibit subsequent ligand binding. Receptors preincubated with ligands, however, will be normally stained with the antibody despite ligand binding. There clearly is a structural model which is compatible with this observation, i.e. an epitope which will form a sort of gate after antibody binding which does not allow the ligand to enter a groove within the receptor. However, we would prefer if we can cite a reference for a similar observation. Can anybody help us with such a reference? Thanks for any help on this subject.
