Please my Dear friend!

Try to avoid Rh123 staining which is often giving artefact of measurement
Better use DiC6(3) in between 1nm and 5 nm in order to avoid any cytotoxic
effects or quenching; Take care to use contro with FCCP or mClCCP (Sigma)
that you will have to calibrate for collapsing the mitochondrial membrane
potential depending of you cell culture medium (depending essentially of
the serum added).
Or use JC-1 red fluorescence in the same range of concentration after
having flitrate the solution to discarb all the red aggregate.
