Dear Flowers, One month ago I posted a question about CD10 staning of lymph node GC cells and follicular lymphoma. My question was: One of our pathologists complained about our CD10 flow results of lymph node biopsies and fine needle aspirates, because they don't match with immuohistochemical staining which shows positivity of lymph nodes germinal center cells and follicular lymphoma while the flow shows negativity or very dim stain although this antibody works well in B-cell acut lymphoid leukaemia. I got many helpful answers to my question. Now it seems that the question is answered fully and based on your suggestions I think I've found the solution: CD10 works fine even at low expression levels if the antibody is conjugated with PE instead of FITC. I've compared CD10-FITC and CD10-PE in approximately 10 experiments working with lymph node fine needle aspirate samples. Two examples below illustrate the dramatic difference according to the FITC or PE conjugation: 1. Reactive lymph node, follicular hyperplasia: <File attached: CD10_FITC_PE_1.jpg> 2. Follicular lymphoma: <File attached: CD10_FITC_PE_2.jpg> Thanks again for your help! Pal Jakso University of Pecs Faculty of Medicine Dept. of Pathology 7643, Hungary 12. Szigeti str.
