Dear Michal, It has been published and we have also seen it in our own research that in certain cases CD8 negatively influenced tetramer staining, it could therefore well be that the same happens when using CD3. We see no effect if we first incubate with the tetramer and subsequently with a monoclonal antibody. On the other hand if you use CD3 for activation, it is known that CD3 down-regulates TCR and thus tetramer staining.
