Jim, A very effective do-it-yourself approach is the one of the Alexa Fluor dyes from Molecular Probes. You can select the dye that is most compatible with your red laser (HeNe to Kr) and filters. Conjugations are very easy to do and give stable products. We've found that even the Alexa Fluor 680 is excited sufficiently well with a 17mW HeNe laser at 633nm on the LSR to give a good signal with highly expressed surface antigens.
