Summary: how to clean the sample injection tube

More than 30 kind people replied this issue. Sorry, I cannot write back the all mails. However, many thanks to all of them.

Since this could be a common problem, I just summarize the suggestion in brief.

1.     Try to run warm or hot bleach (10-50%) from the sample port, and then rinse with water.

2.     Two replied the 0.1N NaOH is useful (please see previous discussion).

3.     Use stylette or clean wire to clean the sample injection tube. You can get it by: purchasing from BD, or bothering the BD engineers very often and they could give you one. You could get one from the Tritech engineer inc, and the web site is www.tritechinc.com.

4.     Sonicate the sample injection tube, 20-30 min should be enough.

5.     Vaccum the tube by syringe (5 ¡V 50 ml?). The winged-needle (or butterfly needle) with rubber tube and connector fits the tube well.

6.     Call BD service.

There is a very good troubleshooting flow-chart of FACS machine and the procedure to remove and restore the sample injection tube (with nice pictures).

After removing the sample injection tube, as long as you don't bend the upper metal portion (the shorter piece), you should not need to recalibrate the system. The most important thing is to clean the machine well every time. However, when something happens, hope this is helpful.
