I know this question has been asked before, but when I searched the archives, I found only the question and not the answer. We've used Indo with cells in suspension, are there any hints for using it with adherent cells? Current protocols says you can, but gives no advice as to how. What's the best way to get them into suspension? I have an investigator who found a published protocol which suggested to load the cells in the flask, treat with the agent they're suspecting causes calcium release, then trypsinze. Wouldn't it be better to use the agent after getting the cells into suspension? I would think the change in calcium would happen too quickly to analyze them after trypsinization.
