hi Flowers, Many thanks for the replies re CD30. It appears that CD30 can stain surface membrane and/or golgi bodies . On Friday we had a peripheral blood from someone with ?large anaplastic lymphoma. About 50% of cells were CD45-CD7weak+, and negative for a range of other lymphoid and myeloid markers, including surface CD30. Having just received the suggestion from the mailing list about the golgi bodies (thanks, Ierachmiel), I restained the CD30, relying on the FACSlyse and fixative to permeabilise the already stained cells. Half of the cells stained positive for intracellular CD30, so it looks like we'll be incorporating a cytoplasmic CD30 stain into our panel, or alternatively restaining after lysis and fixation. many thanks, people, what would I do without you? beth Beth Rees Flow Cytometry Lab Pathology Dept Royal Hobart Hospital Hobart, Tasmania Australia ph. 03 6222 8913
