Hi Joseph, I guess you want to see some subsets of memory/activated and naive T cells (by default), is that right? I suggest the following tubes if you can have four colors a) One tube with CD3/CD4/CD45RO/CD62L b) One tube with CD3/CD8/CD45RO/CD62L Then you will only see T cell subsets If you can have only three colors a) One tube with CD8/CD45RO/CD62L b) One tube with CD4/CD45RO/CD62L In this combination, there is the possibility that you can see other cells that can carry CD45RO. I suggest to do the CD8 because the CD8 part of the T cells have not been so deeply evaluated.
