Dear flowers:
I am working on ApoE knockout mice that develop atherosclerosis and
would like compare the realtive percentage of macrophages in mouse
splenocytes and peripheral blood samples and also measure cytokine
levels, specifically TNF alpha and IL-6, in activated macrophages. What
are the good Macrophage markers available for FACS. We tried CD107b as
a Mac tag but couldn't detect anything, possibly due to sheer
inexperience. For those of you folks who are macrophage and cytokine
savvy...please help! We would also like to look at ox-LDL scavenger
receptors on these macs. Could my macs be sticking to the plastic tubes
I use for flow. Are they any particular staining techniques required? I
used the Pharmingen staining protocol for whole blood. I would really
appreciate some advice on how to proceed into this sticky realm. We use
a dual laser FACS Calibur. Literature references, protocols and
suggestions are welcome.
Thanks

Yasmin Marikar
